ICU> When the ISU instance is removed for a server, other interfaces were not properly detecting that and were behaving as though there was still an ISU instance for that server.

There are documented known issues related to the slow ICU loading times as you mentioned: ICU takes a minute to load on the ISU part & ICU> When the ISU instance is removed for a server, other interfaces were not properly detecting that and were behaving as though there was still an ISU instance for that server; however, there are no plans at this time to resolve the issue.

Unfortunately, it is not possible to disable this behavior in which ICU searches for ISU instances on all of the interface machines that are listed in the Module Database under %OSI\Interfaces\ on each PI Server it is configured to look at. If you are experiencing long loading times, it may be due to ICU scanning multiple interfaces from a number of different servers. You can minimize the impact and make it load data from a select list of PI servers (if you have multiple configured) in ICU>Tools>Options.

Even if all ISU instances are removed from the environment, ICU still checks for ISU on startup from all known collectives, even if the default server is selected and no ISU instances exist that buffer data to that PI Server.
